Effects of Endotoxin treatment on Glucose Production in the Isolated Perfused Rat Liver Preparation

Abstract
|
|
|
Endotoxicosis disturbs glucose metabolism such that the transition to irreversible shock is characterized by progressive hypoglycemia. The current study investigated the effect of endotoxin treatment on glycogenolysis and gluconeogenesis is isolated livers perfused with oxygenated Krebs-Ringer bicarbonate solution. Glucose output from the liver was continuously recorded using a Glucose Analyzer (YSI Model 26 Monitor) and Polygraph (Grass Model 7). The linear rate of glucose production was determined during a 20 min period of perfusion with 10 mM lactate as a substrate for gluconeogenesis and ATP as a stimulant for glycogenolysis. Hormonal responsivity was also assesed by addition of glucagon into the perfusate.
The Key findings were:
1) Rats exposed to high endotoxin progressed to hypoglycemia within 5 hours.
2) Endotoxin treatment reduced basal and ATP-induced glycogenolysis.
3) Normal and glucagon-stimulated gluconeogenesis were inhibited by endotoxin treatment.
In conclusion, glycogenolysis and gluconeogenesis and depressed in late stage endotoxicosis and the glucagon responsivity is reduced.
